Pearce, L. E. (University of Otago, Dunedin, New Zealand), and J. S. Loutit. Biochemical and genetic grouping of isoleucine-valine mutants of Pseudomonas aeruginosa. J. Bacteriol. 89:58-63. 1965.-Isoleucine- and valine-requiring mutants of Pseudomonas aeruginosa, induced with ultraviolet and ethyl-methane-sulfonate, were isolated. The feeding properties and growth requirements of 56 of these mutants were examined, and evidence was found for at least four phenotypic groups, A, B, C, and D, controlling the biosynthesis of isoleucine and valine. Preliminary genetic mapping by use of bacteriophage-mediated transduction showed linkage between the mutant loci within groups A, B, and C. There was also evidence of linkage between the genes controlling groups A and B and some slight evidence of linkage between the other groups. There was insufficient evidence to suggest the arrangement of the genes.

Regulation of isoleucine-valine biosynthesis in Pseudomonas aeruginosa. I. Characterisation and mapping of mutants.
ENE loci controlling enzymes in certain biosynthetic pathways in Escherichia Gcoli and Salmonella typhiznurium have been shown to be clustered on the chromosome (AMES and MARTIN 1964; DEMEREC 1964). The gene loci in some of these clusters are coordinately controlled from an operator locus situated at one end of the cluster. Such a unit o i coordinate expression is called an operon (JACOB and MO...
متن کاملCharacterization of the mmsAB operon of Pseudomonas aeruginosa PAO encoding methylmalonate-semialdehyde dehydrogenase and 3-hydroxyisobutyrate dehydrogenase.
A 5417-base pair (bp) region of Pseudomonas aeruginosa PAO chromosomal DNA containing the mmsAB operon and an upstream regulatory gene (mmsR) has been cloned and characterized. The operon contains two structural genes involved in valine metabolism: mmsA, which encodes methylmalonate-semialdehyde dehydrogenase; and mmsB, which encodes 3-hydroxyisobutyrate dehydrogenase. mmsA and mmsB share the s...
متن کاملGenetic Blocks in the Isoleucine-valine Pathway of Neurospora Crassa.
A group of mutants of Neurospora crassa which require both isoleucine and valine have been described in a series of previous communications from this laboratory (WAGNER, SOMERS and BERGQUIST 1960; WAGNER, KIRITANI and BERGQUIST 1962; and KIRITANI 1962). These mutants were all found to be closely linked between the linkage group V markers Zys-1 (lysine-1, 33933) and sp (spray) within a segment a...
متن کاملAmino acid substitution in an amidase produced by an acetanilide-utilizing mutant of Pseudomonas aeruginosa.
Mutants of Pseudomonas aeruginosa were isolated which grew with acetanilide (N-phenylacetamide) as the sole carbon source for growth. The mutants could also use acetanilide as a nitrogen source but grew better in the presence of ammonium salts. The acetanilide-utilizing mutants produced amidases which differed in substrate specificity and electrophoretic mobility from the wild-type A amidase an...
متن کاملDifference in sodium requirement of branched chain amino acid carrier between Pseudomonas aeruginosa PAO and PML strains is due to substitution of an amino acid at position 292.
Sodium dependence of leucine transport, a measure of the Na+-coupled leucine-isoleucine-valine II (LIV-II) transport system in Pseudomonas aeruginosa, was compared between two wild-type strains, PAO and PML. The leucine transport activity was saturated at 0.1 mM NaCl for PAO and at 5.0 mM for PML. From kinetics experiments, the apparent Km value for Na+ with respect to leucine transport was est...
